Article Alterations in Niban Gene Expression as a Response to Stress Conditions in 3t3-L1 Adipocytes(Springer, 2020) Cevik, Mehtap; Gunduz, Meliha Koldemir; Deliorman, Gokce; Susleyici, BelginAdipocyte death is important in obesity development. Understanding and prevention of adipocyte deaths may be a molecular approach in the treatment. In the study, we aimed to understand role of Niban gene, which acts as an anti-apoptotic molecule as a response to stress conditions, in adipocytes. 3T3-L1 adipocytes were treated with different doses of linoleic acid, hydrogen peroxide and ethanol; and proliferation of the cells examined with real time monitoring iCELLingence system. Gene expression levels were measured by q-PCR. As a response to 24h 480 mu M linoleic acid treatment, Niban gene expression was found to be higher than control group (p = 0.008), whereas 24 h 90 mM ethanol treatment was determined to be lower than control group (p = 0.008). The highest value of Niban gene expression among H2O2 treatment groups was detected in 4h 600 mu M H2O2 in comparison to control group (p = 0.008). To understand role of Niban in adipogenesis, Niban gene expressions were compared between pre-adipocytes and advanced fat accumulated adipocytes and determined to be significantly different (p = 0.042). Our results suggest that Niban might be involved in stress response process in adipocytes. Early diagnosis is vital, but there are some barriers such as economic, cultural, and personal factors. Considering the disadvantages of radiological imaging techniques or serological analysis methods used in cancer diagnosis, such as being expensive, requiring expertise, and being time-consuming, there is a need to develop faster, more reliable, and cost-effective diagnostic methods for use in cancer diagnosis.Exosomes, which are responsible for intercellular communication with sizes ranging from 30-120 nm, are naturally produced biological nanoparticles. Thanks to the cargo contents they carry, they are a potential biomarker to be used in the diagnosis of cancer. Exosomes, defined as extracellular vesicles of endosomal origin, are effective in cancer growth, progression, metastasis, and drug resistance, and changes in microenvironmental conditions during tumor development change exosome secretion. Due to their high cellular activity, tumor cells produce much higher exosomes than healthy cells. Therefore, it is known that the number of exosomes in body fluids is significantly rich compared to other cells and can act as a stand-alone diagnostic biomarker. Cancer-derived exosomes have received great attention in recent years for the early detection of cancer and the evaluation of therapeutic response.In this article, the content, properties, and differences of exosomes detected in common types of cancer (lung, liver, pancreas, ovaries, breast, colorectal), which are the leading causes of cancer-related deaths, are reviewed. We also discuss the potential utility of exosome contents as a biomarker for early detection, which is known to be important in targeted cancer therapy.Article Comparing Cognitive Behavioral Therapy and Ericksonian Hypnotherapy for Obsessive–Compulsive Disorder: A Randomized Controlled Trial(Routledge, 2026) Çınaroğlu M.; Yılmazer E.; Odabaşı C.; Ülker S.V.; Tan S.; Hızlı Sayar G.Obsessive–Compulsive Disorder (OCD) is a chronic condition that often responds well to Cognitive Behavioral Therapy (CBT), though many patients fail to achieve full remission. Ericksonian Hypnotherapy (EH) has been proposed as a promising alternative. This randomized controlled trial compared the efficacy of CBT and EH against a waitlist control in 99 adults with OCD. Participants received 12 weekly online therapy sessions. Outcomes were assessed at baseline, mid-treatment, and post-treatment using the Padua Inventory-Revised, Yale–Brown Obsessive Compulsive Scale–Self-Report (Y-BOCS-SR), and Beck Anxiety Inventory (BAI). Both CBT and EH led to large, statistically and clinically significant reductions in OCD symptoms compared to waitlist. CBT was more effective for compulsive behaviors such as washing, while EH produced greater reductions in obsessive rumination and general anxiety. No serious adverse events occurred. These findings suggest that EH is a viable and comparably effective treatment to CBT, with distinct therapeutic benefits. EH may offer a particularly useful option for patients with obsession-dominant symptom profiles or comorbid anxiety. © 2026 International Journal of Clinical and Experimental Hypnosis.Article Comparing Cognitive Behavioral Therapy and Ericksonian Hypnotherapy for Subclinical Depression and Anxiety: A Randomized Controlled Trial(Routledge Journals, Taylor & Francis Ltd, 2025) Cinaroglu, Metin; Yilmazer, Eda; Odabasi, Cemre; Ulker, Selami Varol; Sayar, Gokben HizliThis randomized controlled trial examined the effectiveness of Cognitive Behavioral Therapy (CBT) and Ericksonian Hypnotherapy (EH) in reducing symptoms of depression and anxiety among individuals with subclinical levels of distress. A total of 150 participants were screened, and 45 eligible participants were randomized into three groups: CBT (n = 15), EH (n = 15), and a waitlist control group (n = 15). Interventions consisted of 12 weekly sessions, with assessments conducted at baseline, mid-intervention, and post-intervention using the Beck Depression Inventory-II (BDI-II) and Beck Anxiety Inventory (BAI). Both CBT and EH significantly reduced symptoms of depression and anxiety compared to the control group, with no significant differences in efficacy between the two interventions. CBT's structured approach demonstrated consistent improvements across both depression and anxiety, while EH showed a slightly faster reduction in anxiety symptoms at mid-intervention, highlighting its potential for addressing somatic and cognitive dimensions of anxiety. This study extends previous findings by focusing on a non-clinical population, emphasizing the potential of these interventions as early strategies for preventing symptom escalation. Unlike prior work primarily targeting diagnosed populations, this research underscores the applicability of CBT and EH in addressing subthreshold distress. Limitations include a small sample size, lack of follow-up assessments, and reliance on self-reported measures. Future research should explore long-term outcomes, larger samples, and the integration of CBT and EH. Constant advancement in diagnostic, medical, and chemical applications has been demonstrated in several platforms and tools. In this study, the analytical and FEA of the microcantilever used in biomolecular analyses were compared with the experimental analysis results.Methods In this study, MITF antigen, which is a melanoma biomarker, and anti-MITF antibody (D5) were selected as biomolecules. A MEMS-type microcantilever biosensor was designed by functionalizing the AFM cantilever by utilizing the specific interaction dynamics and intermolecular binding ability between both molecules. Surface functionalization of cantilever micro biosensors was performed by using FEA. The stress that will occur as a result of the interactions between the MITF-D5 has been determined from the deviation in the resonant frequency of the cantilever.Results It has been found that the simulation results are supported by analytical calculations and experimental results.Conclusion The fact that the results of the simulation study overlap with the experimental and mathematical results allows us to get much cheaper and faster answers compared to expensive and time-consuming experimental approaches.Article Discriminant Analysis Using MRI Asymmetry Indices and Cognitive Scores of Women with Temporal Lobe Epilepsy or Schizophrenia(Springer Science and Business Media Deutschland GmbH, 2024) Fırat, Z.; Er, F.; Noyan, H.; Ekinci, G.; Üçok, A.; Ulǔg, A.M.; Aktekin, B.Purpose: This study aims to assess the diagnostic power of brain asymmetry indices and neuropsychological tests for differentiating mesial temporal lobe epilepsy (MTLE) and schizophrenia (SCZ). Methods: We studied a total of 39 women including 13 MTLE, 13 SCZ, and 13 healthy individuals (HC). A neuropsychological test battery (NPT) was administered and scored by an experienced neuropsychologist, and NeuroQuant (CorTechs Labs Inc., San Diego, California) software was used to calculate brain asymmetry indices (ASI) for 71 different anatomical regions of all participants based on their 3D T1 MR imaging scans. Results: Asymmetry indices measured from 10 regions showed statistically significant differences between the three groups. In this study, a multi-class linear discriminant analysis (LDA) model was built based on a total of fifteen variables composed of the most five significantly informative NPT scores and ten significant asymmetry indices, and the model achieved an accuracy of 87.2%. In pairwise classification, the accuracy for distinguishing MTLE from either SCZ or HC was 94.8%, while the accuracy for distinguishing SCZ from either MTLE or HC was 92.3%. Conclusion: The ability to differentiate MTLE from SCZ using neuroradiological and neuropsychological biomarkers, even within a limited patient cohort, could make a substantial contribution to research in larger patient groups using different machine learning techniques. © The Author(s), under exclusive licence to Springer-Verlag GmbH Germany, part of Springer Nature 2024.Article Effects of Ericksonian Hypnotherapy Versus Cognitive Behavioral Therapy on Prolonged Grief Among University Students: A Randomized Clinical Trial(Routledge Journals, Taylor & Francis Ltd, 2025) Cinaroglu, Metin; Yilmazer, Eda; Ulker, Selami Varol; Cinar, FadimeThis study evaluates the comparative effectiveness of Ericksonian Hypnotherapy (EH) and Cognitive Behavioral Therapy (CBT) in addressing prolonged grief among university students in Istanbul. Transitioning through critical developmental phases, 39 students who had experienced significant loss and demonstrated symptoms of prolonged grief participated in this randomized controlled trial. They were divided equally into three groups: those receiving EH, those undergoing CBT, and a control group placed on a waiting list. Assessments utilized the Prolonged Grief Scale and the Beck Depression Inventory to measure the interventions' impact on grief. Findings revealed significant improvements in both EH and CBT groups compared to the control, with EH showing marginally higher effectiveness in reducing grief symptoms. This research underlines EH's potential as a culturally adaptable and effective treatment for prolonged grief in a diverse academic setting, advocating for its broader application and further exploration across various populations.Article The Effects of Glipizide on DNA Damage and Nuclear Transport in Differentiated 3T3-L1 Adipocytes(Springer, 2022) Cevik, Mehtap; Caker, Selen; Deliorman, Gokce; Cagatay, Penbe; Gunduz, Meliha Koldemir; Susleyici, BelginBackground Despite commonly use for treatment of type II diabetes, possible effects of glipizide on nuclear transport and DNA damage in cells are unknown. Since clinical response of glipizide may change with aging, the aim of the study was to investigate the effect of glipizide by comparing mature and senescent adipocytes. Methods and results The effects of glipizide were investigated in 3T3-L1 adipocytes. Effective and lethal doses were determined by real-time monitoring iCELLigence system. Comet assay was performed to determine DNA damage and quantitative PCR was conducted to detect gene expression levels. RAN expressions were found to be up regulated in mature 180 mu M glipizide treated adipocytes compared to control group (p < 0.05); whereas down regulated in senescent 180 mu M glipizide treated adipocytes compared to their control adipocytes (p < 0.05). Olive Tail Moment values were significantly higher in mature 180 mu M glipizide treated adipocytes (MTG) and senescent 180 mu M glipizide treated adipocytes (STG) comparing their untreated controls (p < 0.001 and p < 0.001 respectively). Also class 5 comets that shows severe DNA damage were found to be higher in both MTG and STG groups than their controls (p < 0.001 and p < 0.001, respectively). OTM values were higher in STG than MTG (p < 0.001). Conclusions This is the first study that reports glipizide caused DNA damage increasing with senescence in adipocytes. As a response to glipizide treatment Ran gene expression increased in mature; and decreased in senescent adipocytes. Further studies are needed to reveal the effect of glipizide on DNA and nuclear interactions in molecular level.Article Ethical Leadership and Perceived Organizational Fairness: A Moderated Mediation Model of Moral Identity and Psychological Contract Breach(Routledge Journals, Taylor & Francis Ltd, 2025) Aktas, Burak Nedim; Gunalan, Mustafa; Akcin, Kultigin; Erat, SerhatThis study identifies when and how ethical leadership is linked to employees' perceptions of organizational fairness. We propose and test a mechanism in which followers' moral identity explains this link, and the state of the psychological contract sets its boundaries, highlighting a novel, process-focused view of fairness formation. Using a cross-sectional survey of 306 white-collar employees in T & uuml;rkiye's industrial sector, we estimated a moderated-mediation model with partial least squares structural equation modeling and bootstrap inference. Results revealed that ethical leadership related to perceived organizational fairness indirectly through the symbolization (outward expression) facet of moral identity, whereas internalization (inward conviction) was unrelated. This indirect link emerged only when employees perceived that promised obligations were honored, indicating that psychological-contract fulfillment enables ethical signals to be read as fair. The findings clarify that visible moral behavior matters more than inward conviction for fairness judgments and that relational integrity is a precondition for leader influence. We discuss implications for theory and for building fair climates by making ethics visible and keeping promises.Article In Vitro Determination of Antileshmanial Activities of Benzimidazolium Derivatives on L. Major Promastigotes and Amastigotes(Springer Int Publ Ag, 2023) Kelleci, Kubra; Golebatmaz, EdaPurpose Leishmaniasis is a serious public health problem infecting millions of people worldwide. An effective and reliable treatment method to be used in the treatment of the disease has not been developed yet. Methods In this article, the anti-leishmanial activities of two benzimidazolium derivatives (B.A and B.B) against Leishmania major promastigotes and amastigotes, which are known to cause cutaneous leishmaniasis, were investigated for the first time. The immunostimulatory activity of the developed formulations was determined using the J774 murine macrophage cell line. Results B.A and B.B compounds were found to have a much higher cytotoxic effect than Amphotericin B (IC50 value 0.75 mu M +/- 0.03), which is used as the reference drug. The IC50 value was determined as 2.02 mu M +/- 0.52 for B.A and 1.83 mu M +/- 0.71 for B.B in Leishmania promastigotes. In addition, IC50 values of B. A and B.B Leishmania amastigotes were found to be 1.01 mu M and 0.67 mu M, respectively. It was found that B.B was 81.12 times more selective than Amphotericin B and showed the highest selectivity against L. major promastigotes (359.09) and amastigotes (980.80). Considering the selectivity indices (SI) of B.A and B.B, both compounds tested are more promising than Amphotericin B. Conclusion The results showed that benzimidazolium derivatives have anti-leishmanial potential against L. major, which is the causative agent of cutaneous leishmaniasis. Thus, we can say that the obtained results will help the development of effective and safe antileishmanial drug formulations against cutaneous leishmaniasis.Article Investigation of Dpyd, Mthfr and Tyms Polymorphisms on 5-Fluorouracil Related Toxicities in Colorectal Cancer(Newlands Press Ltd, 2022) Cevik, M.; Namal, E.; Şener, N.D.; Köksal, U.I.; Çaǧatay, P.; Deliorman, G.; Çiftçi, C.Aim: To investigate the association of DPYD, MTHFR and TYMS polymorphisms on 5-fluorouracil (5-FU) related toxicities and patient survival. Materials & methods: A total of 103 colorectal cancer patients prescribed 5-FU were included in the study. Genotyping was conducted for several DPYD, MTHFR and TYMS polymorphisms using a microarray analyzer. Results: DPYD 496A>G polymorphism was found to be significantly associated with 5-FU related grade 0-2, but not severe toxicities (p = 0.02). Furthermore, patients with DPYD 85TC and CC genotypes had longer progression and overall survival times compared to TT genotypes in our study group (log rank = 6.60; p = 0.01 and log rank = 4.40; p = 0.04, respectively). Conclusion: According to our results, DPYD 496AG and GG genotypes might be protective against severe adverse events compared to the AA genotype. Another DPYD polymorphism, 85T>C, may be useful in colorectal cancer prognosis. Further studies for both polymorphisms should be conducted in larger populations to achieve accurate results. © 2022 Future Medicine Ltd.Article Menstrual Cycle-Related Changes in Women with Schizophrenia: A Resting-State fMRI Study(Karger, 2022) Noyan, Handan; Hamamci, Andac; Firat, Zeynep; Sarsilmaz, Aysegul; Ucok, AlpIntroduction: Different influences of ovarian hormones in schizophrenia (SCZ) have been reported, but no study to date has assessed their effects on the brain dynamics at rest. The present study aimed to examine the hormonal and clinical changes related to the menstrual cycle and alterations in the resting-state functional connectivity (RS-FC) depending on cycle phase and/or hormonal fluctuations in SCZ. Method: This study was conducted based on both between- and within-subject experimental designs, including 13 clinically stable female patients with SCZ (32 +/- 7.7 years) and 13 healthy women (30 +/- 7.3 years). RS-functional magnetic resonance imaging (fMRI) scanning, as well as hormonal and clinical assessments, was applied to each participant twice during two cycle phases: early follicular and mid-luteal. Results: A difference in mid-luteal progesterone levels was found between groups, with a large effect size (Cohen's d) of 0.8 (p < 0.05). Also, the estradiol levels negatively correlated with the negative symptom severity of the patients during their mid-luteal phase. In the patients, estrogen positively correlated with the auditory network connectivity in the left amygdala during the early follicular phase. In the controls, progesterone had positive correlations with the connectivity of the posterior default mode and the left frontoparietal networks in the bilateral precuneus during the early follicular phase and had a negative correlation with the executive control network connectivity in the mid-luteal phase. Conclusion: The present study showed hormonal differences between groups and suggested that the levels of cycle-dependent hormones might be associated with the changes in clinical symptom severity and the RS-FC in the groups. Therefore, drugs targeting mTOR have been developed. Although mTOR inhibitors rapamycin and rapamycin rapalogs (everolimus, rapamycin, temsirolimus, deforolimus, etc.) and new generation mTOR inhibitors (Rapalink, Dual PI3K/mTOR inhibitors, etc.) are used in cancer treatments, mTOR resistance mechanisms may inhibit the efficacy of these drugs. Therefore, new inhibition approaches are developed. Although these new inhibition approaches have not been widely investigated in cancer treatment, the use of nanoparticles has been evaluated as a new treatment option in a few types of cancer.This review outlines the functions of mTOR in the cancer process, its resistance mechanisms, and the efficiency of mTOR inhibitors in cancer treatment. Furthermore, it discusses the next-generation mTOR inhibitors and inhibition strategies created using nanoparticles.Since mTOR resistance mechanisms prevent the effects of mTOR inhibitors used in cancer treatments, new inhibition strategies should be developed. Inhibition approaches are created using nanoparticles, and one of them offers a promising treatment option with evidence supporting its effectiveness.Article Of the Manuscript: Predictive Value of Renal Parenchyma and Pelvic Urine Density on Preoperative Non-Contrast CT in Systemic Inflammatory Response Syndrome (SIRS) After Semi-Rigid Ureteroscopy(Springer, 2025) Ozsoy, Emrah; Kutluhan, Musab Ali; Tokuc, Emre; Kayar, Ridvan; Demir, Samet; Meric, Kaan; Ozturk, Metin IshakTo evaluate the predictive value of preoperative NCCT (Non-Contrast Computed Tomography) measured renal parenchyma and pelvic urine densities for the development of systemic inflammatory response syndrome (SIRS) following URS. 1262 patients underwent semi-rigid URS due to ureterolithiasis between January 2008 and August 2023 were screened and 286 patients were included. Patient demographics and NCCT findings (stone size, location, density, hydronephrosis, renal parenchymal density, pelvic urine density) were collected retrospectively. Patients were categorized into two groups based on SIRS development after URS. Pelvic urine and parenchyma densities of the obstructed kidney were compared. A multivariate logistic regression analysis was performed to identify independent predictors of SIRS. The median obstructed kidney cortex mean density was significantly lower in the SIRS (+) group compared to the SIRS (-) group (26.67 [22.33-29.50] vs. 33.33 [27.24-37.75]). Obstructed kidney medulla mean density was also significantly lower in patients with SIRS (24.00 [22.33-27.50] vs. 31.50 [26.24-36.75], p = 0.001). Pelvic urine density was significantly higher in the SIRS-positive group. Stone density (OR = 1.35, % 95 CI: 1.09-1.67, p = 0.006), proximal ureteral location (OR = 5.21, % 95 CI: 1.84-14.7, p = 0.002), cortex mean density (OR = 0.91, % 95 CI: 0.87-0.95, p = 0.001) and pelvic urine density (OR = 1.07, % 95 CI: 1.02-1.12, p = 0.010) were stated as significant predictors. Renal parenchyma and pelvic urine density on pre-operative NCCT can predict SIRS after semi-rigid URS.Article Regenerative Therapy Approaches and Encountered Problems in Sensorineural Hearing Loss(Bentham Science Publ Ltd, 2023) Kelleci, Kuebra; Golebetmaz, EdaHearing loss is one of the most important public health matters worldwide, severely affecting people's social, psychological, and cognitive development. The perception of sound, movement, and balance in vertebrates depends on a special sensory organ called the cochlea, which contains hair cells and supporting cells in the inner ear. Genetic factors, epigenetics, the use of ototoxic drugs (some antibiotics and chemotherapeutics), noise, infections, or even aging can cause loss of hair cells and their related primary neurons, leading to sensorineural hearing loss. Although a sensorineural hearing loss, also known as permanent hearing loss, is treated with hearing aids and cochlear implants, treatment methods are limited. Since even the best implant cannot exhibit the characteristics of the original ear, the permanent sensory deficit will be permanent. For this reason, it has become important to develop regenerative treatment methods to regenerate and replace lost or damaged hair cells and neurons. Developments in stem cell technology have led to promising studies in regenerating damaged/lost hair cells or neurons with endogenous or exogenous cell-based therapies. Epigenetic mechanisms can turn hearing-related genes on and off and determine which proteins to copy. In addition, due to gene silencing, gene replacement, and CRISPR/CAS9 technology, gene therapy methods have accelerated, and studies have been carried out to treat dominant and recessive mutations that cause genetic-induced hearing loss or increase hair cell regeneration. In this paper, potential gene therapy and stem cell applications in the acquisition of cochlear function, which causes sensorineural hearing loss, and the difficulties encountered in these applications are compiled from a bioengineering perspective.Article Results in Kidney Transplant Recipients from Living Donors 75 Years of Age or Older(Elsevier Science Inc, 2024) Cicek, Sevim Nuran Kuslu; Huseynov, Amil; Tuncer, MuratThe increasing age of patients receiving renal replacement therapy (RRT) in Turkey, coupled with a shortage of kidney donors, has led to longer waiting times for transplants and an escalation in mortality rates. This retrospective study aimed to assess the effect on transplant outcomes of accepting kidneys from donors >= 70 years of age, given the rising number of older patients in the population. In all, 1400 patients were transplanted with kidneys from donors >50 years, with patient and graft survival as primary endpoints. Our results demonstrated that the most significant risk factors for graft function were recipient age >65 years, male sex, and presence of type 2 diabetes. Moreover, kidneys from donors >= 75 years of age achieved a half-life of 5 years. These findings suggest that donor age does not necessarily correlate with graft failure and that transplantation from older donors could help alleviate the organ shortage. Further research is needed to substantiate these conclusions.Article School-Based Psychosocial and Educational Interventions for Children and Adolescents After the 1999 Marmara Earthquakes in Turkey: A Review on Lessons Learned(Pergamon-Elsevier Science Ltd, 2024) Cihanoglu, Mine; Vatansever, Merve; Erden, GulsenThe massive earthquakes experienced in August and November 1999 affected thousands of people in the Marmara region, the most densely populated and industrialized part of Turkey. The humanitarian and economic cost was so enormous, and these earthquakes have changed the Turkish disaster management system and the Turkish people's approach to disasters. Marmara earthquakes are also considered as a milestone in the provision of psychosocial services for disaster victims. This paper aims to review the psychosocial interventions targeting children, adolescents, and their families after the 1999 earthquakes in Turkey. The progression from initial responses to more organized psychosocial interventions is outlined. Conducting the interventions at schools has ensured that thousands of children, teachers, and parents are reached in the most efficient and effective way possible. The significance of the school context in designing psychosocial interventions is highlighted and implications of the lessons learned for traumatic experiences of children and parents are also explored. It is evaluated that these inferences obtained from the Marmara earthquake in Turkey can be used in disasters around the world.
